چکیده

1. It is shown that free haematins will react with nitric oxide under certain conditions giving compounds with characteristic absorption spectra. 2. The reactions of nitric oxide with both protohaematin and urohaematin were examined. In acid solution each of these haematins changes colour from reddish brown to a bright red in the presence of nitric oxide, and two strong absorption bands appear in the green region ofthe spectrum while the Soret band of the haematin is shifted to a longer wavelength. 3. Urohaematin in alkaline solution reacts similarly with nitric oxide and can be reduced to give a NO-urohaem compound with a single broad absorption band in the visible region. The results obtained with alkaline protohaematin are less clear-cut. 4. The absorption spectra of the NO-compounds ofprotoanduro-haematin under various conditions have been determined. 5. No spectroscopic evidence was obtained for any reaction between fluoride, azide or sulphide with free haematins, although these substances all form well-defined compounds with methaemoglobin and other haemoproteins. 6. The reactions of different substances with free iron-porphyrins were compared with those given with haemoproteins. Of all the substances examined only nitric oxide and the cyanide ion are capable of co-ordinating with the bivalent and tervalent iron of haem and haematin respectively, both when they are free and when they form the prosthetic groups of haemoproteins.

Reactivities of hydroxylamine and sodium bisulphite with carbonyl-containing haems and with the prosthetic groups of the erythrocyte green haemoproteins.

The reactivities of alkaline NH(2)OH and neutral NaHSO(3) with carbonyl and olefinic groups conjugated with the tetrapyrrole nucleus of haems were studied. The reactions were carried out with 2-3nmol of haem a, spirographis haem, isospirographis haem, 2,4-diacetyldeuterohaem and protohaem. Vinyl side chains were found to be insensitive to the chemical action of both alkaline NH(2)OH and neutral...

CO-reacting haemoproteins of neutrophils: evidence for cytochrome b-245 and myeloperoxidase as potential oxidases during the respiratory burst.

Room temperature, CO-difference spectra of intact rat polymorphonuclear leucocytes (neutrophils) revealed the presence of a number of CO-binding haemoproteins. Absorption maxima at 413, 540 and 570 nm were attributed to the CO-complex of cytochrome b-245 whereas an absorption maximum at 595 nm was assigned to the contribution from a myeloperoxidase complex, since an identical absorption maximum...

Photochemical action spectra indicate that cytochrome a/a3 is the predominant haemoprotein terminal oxidase in Acanthamoeba castellanii.

1. Room-temperature CO-reduced minus reduced difference spectra of intact cells of Acanthamoeba castellanii show the presence of CO-reacting haemoproteins in cells from the early-exponential, late-exponential and stationary phases of growth. 2. The relative rates of reaction with CO of the two haemoproteins differ; that of cytochrome a/a3 with CO is complete within 1 min of bubbling with CO, wh...

Diverse structural approaches to haem appropriation by pathogenic bacteria.

The critical need for iron presents a challenge for pathogenic bacteria that must survive in an environment bereft of accessible iron due to a natural low bioavailability and their host's nutritional immunity. Appropriating haem, either direct from host haemoproteins or by secreting haem-scavenging haemophores, is one way pathogenic bacteria can overcome this challenge. After capturing their ta...
